CompanyDocumentnumber：WTUT-WT88Y-W8BBGB-BWYTT-19998CFSE细胞增殖实验准备：20ml预冷5%FBS(HI)-PBS50ml预冷MACSbuffer70um滤网RBClysisLScolumn50ml37度预热PBS50ml37度预热completeculturemedium(RPMI1640+10%HeatInactivatedFBS+10mMHEPES+1mMpenicillinestreptomycin+50mM2-mercaptoethanol)（一）CD3抗体包被取96孔板每孔加入100ul10ug/mlanti-CD3抗体（PBS）密封4度过夜。（二）淋巴细胞获取小鼠的脾脏结摘取于10ml冷5%FBS-PBS中。于70um滤网研磨滤过，500g4度5min离心后弃上清。RBClysis裂红5min，10ml冷5%PBS终止。500g4度5min离心后弃上清，10mlMACSbuffer重悬，计数。留取105个细胞进行FACS。磁珠分选naveCD8a+T细胞cellsuspensionat300×gfor10minutes.Aspiratesupernatantcompletely.cellpelletin400μLofMACSbufferper108totalcells.100μLofNaiveCD8a+TCellBiotin-AntibodyCocktailper108totalcells.wellandincubatefor5minutesintherefrigerator(28°C).Add200μLofMACSbufferper108totalcells.Add200μLofAnti-BiotinMicroBeadsper108totalcells.Add100μLofCD44MicroBeadsper108totalcells.Mixwellandincubateforanadditional10minutesintherefrigerator(28°C).Washcellsbyadding10mlofMACSbufferper108cellsandcentrifugeat300×gfor10minutes.Aspiratesupernatantcompletely.Resuspendupto108cellsin500μLofMACSbuffer.PlaceLScolumninthemagneticfieldofasuitableMACSSeparator.Preparecolumnbyrinsingwith3mLofMACSbuffer.Applycellsuspensionontothecolumn.Collectflow-throughcontainingunlabeledcells,representingtheenrichednaiveCD8a+Tcellfraction.Washcolumnwith3mLofMACSbuffer.Collectunlabeledcellsthatpassthrough,representingtheenrichednaiveCD8a+Tcells,andcombinewiththeflow-throughfromstep3.Determinecellnumber.留取105个细胞进行FACS。（四）CFSE预处理1.Removesupernatantfromthecellpellet.2.AddCellTraceCFSE(1:1000dilutioninPBS)stainingsolutionandgentlyresuspendthecellsto1*106/ml.3.Incubateat37°Cfor20minutes,protectedfromlight.4.Add4volumescompleteculturemediumandmix.5.Incubateat37°Cfor5minutes.6.Pelletthecellsandremovethesupernatant.7.Resuspendthecellpelletinfresh,pre-warmedcompleteculturemediumto1*107/ml.8.均分两份，其中一份加入2ug/mlanti-CD28，一份不加抗体。9.包被板使用PBS洗涤一次，按105-106个细胞／孔梯度接种，培养3天后上机。
