卵母细胞的成熟需要多种信号的调节,cAMPPKC和MAPK对卵母细胞的成熟具
有重要作用.通过体外培养猪的卵丘包裹的卵母细胞(COC)利用蛋白电泳和蛋白免疫
印迹技术,本文研究了猪卵母细胞体外成熟过程中cAMP和PKC对MAPK活性的影响
以及这三种信号通路对猪卵母细胞成熟的调节发现增加cAMP浓度的forskolin和1BMx
可抑制MAPK的活性和猪卵发生GvBD,100.50,25mforskolin或10,5rnMIBMx可
明显抑制MAPK的激活2/aM蛋白磷酸酶抑制剂OA可反转forskolin或IBMx对MAPK
激活的抑制PKC激活荆PMA可强烈抑制MAPK的激活和猪卵发生GVBD,162nM
PMA可完全抑制MAPK激活,l6.2,l62nMPMA可明显抑制MAPK的激活.无生理活
性的PMA类似物4a-pDD对MAPK激活和GVBD无任何影响生理性PKC激活剂diC
可抑制MAPK的激活和猪卵发生GVBD,1000,500pMPMA可明显抑制MAPK的激活.
1gMPKC抑制剂ca1FlhostinC或30nM广谱蛋白激酶抑制剂staurosporine可反转PMA或
diC对MAPK激活的抑制.以上结果揭示cAMP和PKC可负调节MAPK的激活,三者对
于猪卵成熟具有重要调节作用
